- MYB proto-oncogene encodes a transcription factor regulating hematopoietic cell proliferation, differentiation, survival.
- Contains DNA-binding, transactivation, and regulatory domains; controls cell cycle genes, lineage genes.
Fusion effects
- MYB-NFIB:
- t(6;9)(q22-23;p23-24) → delete C-terminal regulatory domain, causing overexpression of MYB targets, promoting oncogenesis.
- Hallmark of adenoid cystic carcinoma (salivary gland, breast, head/neck), dermal cylindroma.
- MYB/MYBL1-QKI:
- Due to chromosome 6q intrachromosomal deletion. → drives tumorigenic gene expression, proliferation.
- Present in the following tumour
