Mutation effects
- Point mutations (Q61R/K/L, G12D/V, G13D/R) impair GTPase activity, lock in GTP-bound state, constitutive activation of downstream effectors.
- Q61 mutations preferred in melanoma due to higher stability/oncometabolite.
